چکیده مقاله:

Objective(s)The key transcriptional regulator Oct4 is one of the self-renewal and differentiation-related factors in cancer stem cells, where it maintains "stemness" state. Cancer stem cells have been identified in a variety of solid malignancies. They are a small population of tumor cells with stem cell characteristics, which are a likely cause of relapse in cancer patients. Due to high incidence, mortality, and recurrence rates of bladder cancer and the necessity of accurate prediction of malignant behavior of the tumors, we evaluated the prognostic value of Oct4 expression in formalin-fixed paraffin-embedded (FFPE) tissues of bladder cancer.Materials and MethodsIn this study, Oct4 expression was evaluated in 52 (FFPE) tissues of bladder cancer. RNA extraction from samples of 30 patients from the archive of Labbafi-Nejad Medical Centre in Tehran was performed and Oct4 expression levels were examined by semi-quantitative RT-PCR. The intracellular distribution of Oct4 protein was also determined by immunohistochemistry (IHC).ResultsThe results revealed a significant correlation between the expression level of Oct4 and the tumors’ grade and stage. A mostly cytoplasmic distribution of Oct4 protein was also confirmed by IHC.ConclusionAll together, our data indicate that the expression level of Oct4 gene is correlated with the clinical and histopathological prognostic indexes of tumors and thus can be considered as a potential prognostic tumor marker.

evaluating the expression of survivin gene as a specific tumor marker for prognosis of bladder cancer

objective: inhibition of apoptosis may favor the onset and progression of cancer. survivin is an inhibitor of apoptosis that has been considered as a potential marker for diagnostic and/or prognostic of bladder cancer. the survivin protein regulates both cell division and cell death and is overexpressed in the vast majority of human cancers. in this study, the expression pattern and potential p...

Is Bax/Bcl-2 Ratio Considered as a Prognostic Marker with Age and Tumor Location in Colorectal Cancer?

Background: Bax and Bcl-2 are the major members of Bcl-2 family whose play a key role in tumor progression or inhibition of intrinsic apoptotic pathway triggered by mitochondrial dysfunction. Therefore, the balance between pro- and anti-apoptotic members of this family can determine the cellular fate. Methods: In this study, the relative level of mRNA expression of Bax and Bcl-2 genes was deter...

Thymidine phosphorylase expression as a prognostic marker for predicting recurrence in primary superficial bladder cancer.

No prognostic marker for predicting recurrence in primary superficial bladder cancer has been established. The aim of this study was to investigate the expression levels of thymidine phosphorylase (TP) by enzyme-linked immunosorbent assay (ELISA) and the immunohistological localization of TP in primary superficial bladder cancer tissue to assess its association with tumor recurrence and stage p...

Circulating Tumor Cells as a Prognostic Marker in Pancreatic Cancer

Pancreatic cancer, with a patient survival rate among the worst of any solid cancer, is the fourth leading cause of cancer-related morbidity and mortality in Western countries (Tjensvoll et al., 2013). It leads to an estimated 227, 000 deaths per year worldwide and the 5 year survival rate is only 4.6% due to its early metastasis (Zhou et al., 2011). Most of the pancreatic cancer patients are a...

mRNA Expression of S100A8 as a Prognostic Marker for Progression of Non-Muscle-Invasive Bladder Cancer

PURPOSE S100A8 is a member of the S100 protein family containing 2EF-hand calcium-binding motifs. S100 proteins are involved in the regulation of a number of cellular processes such as cell cycle progression and differentiation. Altered expression of this protein is associated with various diseases and cancers. The present study aimed to evaluate whether S100A8 has prognostic value for non-musc...
